چکیده

Balish, Edward (Argonne National Laboratory, Argonne, Ill.), and Stanley K. Shapiro. Cystathionine as a precursor of methionine in Escherichai coli and Aerobacter aerogenes. J. Bacteriol. 92:1331-1336. 1966.-Cystathionine has been shown to be a precursor of methionine biosynthesis in Escherichia coli and Aerobacter aerogenes. A double enzyme assay was developed to show the formation of homocysteine from cystathionine. The results obtained support the concept that cystathionine serves as a precursor of methionine via the intermediate formation of homocysteine. The latter compound is methylated by the homocysteine methyltransferase of these microorganisms. Sulfhydryl and keto acid assays were used to demonstrate cystathionase activity. Methionine represses both homocysteine methyltransferase formation and cystathionase formation. However, the presence of methionine in reaction mixtures resulted in product inhibition of homocysteine methyltransferase activity, but not of cystathionase activity.

Biosynthesis in Aerobacter aerogenes

Two pathways for methionine methyl formation, one cobalamindependent and one cobalamin-independent, corresponding to those observed in Escherichia coli PA 15 have been found in Aerobacter aerogenes. An initial difficulty in showing the cobalamin-dependent pathway in cellfree extracts proved to be due to the presence of enzymes which caused the removal of adenosylmethionine, a cofactor required ...

Growth rate of Enterobacteriaceae at elevated temperatures: limitation by methionine.

The effect of elevated temperatures on growth rate was studied in five strains of Enterobacteriaceae. In all the strains tested a shift to the elevated temperature resulted in an immediate decrease in growth rate which was due to limitation in the availability of endogenous methionine. The first biosynthetic enzyme of the methionine pathway-homoserine transsuccinylase-was studied in extracts of...

Response of Aerobacter aerogenes methionine auxotrophs to adenine thiomethyl compounds.

It has been shown that many microorganisms produce thiomethyl adenosine from methionine (Smith and Schlenk, 1952). This prompted biochemical studies on the relationship between methionine and thiomethyl adenosine, which resulted in the discovery of methionyl adenosine as the key intermediate (Catoni, 1952; Schlenk, 1952). Replacement of methionine by thiomethyl adenosine has not been demonstrat...

Methionine-to-cysteine recycling in Klebsiella aerogenes.

In the enteric bacteria Escherichia coli and Salmonella enterica, sulfate is reduced to sulfide and assimilated into the amino acid cysteine; in turn, cysteine provides the sulfur atom for other sulfur-bearing molecules in the cell, including methionine. These organisms cannot use methionine as a sole source of sulfur. Here we report that this constraint is not shared by many other enteric bact...

Preparation and properties of cell-free "formic hydrogenlyase" from escherichia coli.

The available evidence indicates that formic acid is the precursor of the molecular hydrogen produced as an end product of fermentations effected by Escherichia coli, Aerobacter aerogenes, and related organisms. Formate is rapidly decomposed by these bacteria with the formation of H2 and CO2 in a reaction catalyzed by the enzyme or enzyme-complex designated as "formic hydrogenlyase". One of the...
